The insert contains RsaI sites 479 nt and 642 nt from the BamHI cloning site. Restriction digests of the clone give the following sizes (kb): BamHI/HindIII--3.0, 0.67; EcoRI--3.6; HindIII--3.6; BamHI--3.6; PstI--uncut. Mapped to chromosome 16 by linkage with dw (2.7 cM at 95% confidence level). The orientation in the vector is such that aa 47 (the 5' end) is at the BamHI site and aa 269 (the 3' end) is at the HindIII site. Cloned by PCR amplification from cDNA using primers with HindIII and BamHI linkers. The product corresponds to aa 47 to aa 269. |
