The following restriction enzymes do not cut the insert: AatII, BalI, BamHI, BglI, BglII, BstEII, BstXI, HpaI, KpnI, MluI, MstI, NaeI, NarI, NcoI, NdeI, NruI, PaeR7I, PstI, PvuI, SalI, SfiI, SmaI, XbaI, and XhoI. Restriction sites (with nucleotide positions relative to the published sequence) include: AvaII--476; BclI--1548; EcoRI--624; EcoRV--747; HincII--323; HindIII--1262; PvuII--770, 1454; SacI--673, 1833; XmnI--240. Restriction digests of the clone give the following sizes (kb): EcoRI--8.6, 1.9, 0.6; HindIII--6.3, 1.7, 1.3, 1.2, 0.5; BamHI--6.5, 3.2, 1.0; consistent with deposited DNA. The insert contains an EcoRI site at nucleotide 624 so that an EcoRI digest yields 2 fragments from the insert. Contains the complete coding sequence (nt 132-1464). |
