The insert contains internal AccI and XbaI sites. Restriction digests of the clone give the following sizes (kb): SmaI--uncut; BamHI--4.6; EcoRI--4.6; HindIII--4.6; PstI--4.6; EcoRI/HindIII--2.9, 1.7. Linearize the plasmid with EcoRI for in vitro transcription. This is a full-length clone of virus segment S6. The cDNA was tailored to have a 3'-terminal EcoRV site, but this clone lacks the complete sequence. |
