Insert contains the following sites separated by (bp): (EcoRI)- 140- ClaI- 263- BglI- 469- BcnI- 142- MboII- 301- MspI- 72- ClaI- 20- AvaII- 950- RsaI- 198- AccI- 455- (EcoRI). Restriction digests of the clone give the following sizes (kb): XhoI--6.0; BglI--3.5, 1.5, 1.0; HindIII--6.0; EcoRI--3.0, 3.0; AccI--5.4, 0.5. Codominant segregation observed in 9 (PstI RFLP) and 2 (BglII RFLP) families (38 and 10 individuals, respectively). Insert contains most of the sequence (from position 77 to the poly(A) signal) of human thyroid peroxidase. DNA sequence shows 68% homology with the partial porcine thyroid peroxidase sequence. Also detects a VNTR polymorphism best visualized in BglII and EcoRI digests. Requires a last stringent wash (0.1X SSC, 65C). |
