Restriction digests of the clone give the following sizes (kb): PstI/EcoRI--2.7, 0.72, 0.45; PstI--3.9; EcoRI--3.0, 0.8; BamHI--2.8, 1.1; HindIII--3.9. The insert is derived from a conserved region of the gene because it detects 2 HindIII fragments (about 12 kb and 5 kb) in the mouse and 2 HindIII fragments (about 6.8 kb and 6.0 kb) in the Chinese hamster. The 0.72 kb EcoRI fragment detects the RFLP. This probe also detects the same polymorphisms when genomic DNA is cut with SacI, PstI, or ApaI. |
