The insert has an internal HindIII site about 170 bp from the vector HindIII site. The insert also contains DdeI sites (2), TaqI sites (2), Sau3A1 sites (2) , and a RsaI site. Restriction digests of the clone give the following sizes (kb): PstI--3.0, 0.75; EcoRI--3.8; HindIII--3.6, 0.2. Also available from the ATCC are pBYr5 (ATCC 45074) and pBYr5aSP (ATCC 45075) which contain the 0.75 kb PstI fragment. The former is cloned in pUC9 and the latter is also in pSP64, but in the opposite orientation. pBYr5bSP was constructed by random priming. RNA transcribed from the SP6 promoter is complementary to virion sense RNA, i.e. hybridizes to + strand. |
