The insert contains internal sites for AvaII, AluI, BamHI and SmaI. Restriction digests of the clone give the following sizes (kb): HaeIII--0.64, 0.54, 0.48, 0.38, plus smaller fragments; EcoRI--4.7; PstI--4.7; EcoRI/PstI--4.0, 0.74; EcoRI/PvuII--2.5, 2.4. Noninfectious unless released from plasmid by HaeIII digestion (and even then probably noninfectious). Full-length PSTV cDNA with HaeIII ends was cloned into the HindIII site of pBR322 using HindIII linkers. |
