Restriction digests of the clone give the following sizes (kb): BamHI--4.9, 2.8; BglII--7.6; EcoRI--4.9, 2.6; NotI--7.6; SstI--7.0, 0.56. Digestion with EcoRI produces fragments of 5.0 and 2.5 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes the following mutant form: G4F, K5-. The glycine and lysine on residues 4 and 5 of human factor IX were replaced with one phenylalanine. |
