Restriction digests of the clone give the following sizes (kb): BglI--3.2, 1.45, 0.4; EcoRI--2.75, 2.15; XhoI--3.2, 1.8. Plasmid with an XhoI promoter cassette (1.777 kb) containing the wild type lac promoter. The vector component of this construct contains unknown sequence. This should be used only to excise the promoter-containing cassette. |
