Restriction digests of the clone give the following sizes (kb): HindIII--5.4; EcoRI--5.4; PstI--3.8, 1.6. This clone includes the complete coding sequence, some 5' and 3' sequences. This clone detects genomic EcoRI fragments of 11.0 and 4.1 kb. The polymorphic site is located approximately 100 bp downstream of the 3' end of exon 4. Because of the large sizes of the alleles if genomic DNA is digested with NheI alone, detection of the NheI polymorphism is improved with a NheI/SpeI digest. |
