Restriction digests of the clone give the following sizes (kb): BamHI--4.7, 2.7; EcoRI--5.0, 2.5; EcoRV--7.4; PstI--4.7, 2.7; SacI--6.8, 0.5. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-2 and 11-48 of processed human factor VII (Gla domain), with mutations K32E, D33N, A34T, K38T, L39E, replace the corresponding region of factor IX. |
