Restriction digests of the clone give the following sizes (kb): EcoRI--19.0, 15.0, 10.7; BamHI--19.3, 10.7, 4.55, 3.85, 2.6 (approximate); HindIII--19.3, 10.7, 8.0, 4.5, 0.8, 0.58. A HindIII digest gives 4 fragments from the insert (kb): 8.0 kb, 4.5 kb, 0.8 kb, and 0.58 kb. The 8.0 kb fragment is sufficent to express a thermostable DNA polymerase activity and a 60 kDa polypeptide immunologically related to Taq DNA polymerase in Escherichia coli. The 4.5 kb fragment is also available in pFC83, ATCC 67422. |
