The insert contains the following restriction sites (approximate kb from the 5' end): PstI--0.15; BglII--0.16; SpeI--0.97; KpnI--1.18; XhoI--1.37. The plasmid is not stable in E. coli MC1061. Includes nt 19-1632 of the virus, including the complete ORF1 and part of ORF2. Transcription from the T7 promoter gives the sense (+) strand. |
