Restriction digests of the clone give the following sizes (kb): BamHI--4.7, 2.7; BstEII--7.4; EcoRI--5.0, 2.4; NotI--7.4; SacI--7.0, 0.5. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain), with mutations L8F, R9V, P10Q, K32E, D33N, A34T, K38T, L39E, replace the corresponding region of factor IX. |
