Restriction digests of the clone give the following sizes (kb): BamHI--4.4, 3.0; BstEII--7.6; SstI--7.0, 0.5; NotI--7.6; EcoRI--5.0, 2.8. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain), with mutations K32E, K38T, L39E, replace the corresponding region of factor IX. |
