Restriction digests of the clone give the following sizes (kb): BamHI--6.2; BglII--6.2; HindIII--6.2. Can be used, by exploitation of LEU2 selection, for homologous recombination-based insertion into YACs, thus providing one approach to screening YAC libraries. The targets can be repetititive or unique sequences. Enables subsequent transfer into mammalian cells by conferring G418 resistance. Mammalian gene targeting vector designed for replacement or insertional recombination. Suitable for "hit and run" strategies. Inserts must include the promoter and 5' coding sequences of an expressed gene ligated in frame with neo.
