HindIII cuts the insert twice, 7 bp apart. The insert contains internal EcoRI, AvaI, SalI, AccI, HincII, PstI and HindIII sites. Restriction digests of the clone give the following sizes (kb): BamHI--3.1, 1.2; EcoRI--3.1, 0.83, 0.41; HindIII--3.9, 0.38; PstI--3.4, 0.92; XbaI--4.3. Linearize the plasmid with SphI for in vitro transcription. This is a full-length clone of virus segment S10. The cDNA ends were tailored to terminal BamHI sites before cloning into the BamHI site of pGEM-1. |
