The insert contains a KpnI site 0.1 kb from the end of the insert closer to the XhoI site of the vector multiple cloning site. The insert does not contain restriction sites for the following enzymes: ApaI, AatII, BglII, HindIII, PvuII, SacI, XhoI. Restriction digests of the clone give the following sizes (kb): EcoRI--2.9 (doublet); KpnI--3.1, 2.7; PstI--uncut; BglI--2.4, 2.1, 1.3; SalI--uncut. The 1.8 kb EcoRI fragment (pE1.8, ATCC 65264/65265) is immediately adjacent to and downstream from a 2.8 kb EcoRI fragment (pE2.8, ATCC 65260/65261). Contains repeated sequences which must be saturated with sheared total human placental DNA before filter hybridization to remove background. |
