Restriction digests of the clone give the following sizes (kb): HindIII--15.7, 9.5, 4.8, 3.6, 3.2, 2.6; EcoRI--12.8, 6.7, 5.5, 5.1, 3.6, 2.9; BamHI--19.9, 12.0, 7.3, 4.6. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The BamHI fragment is one of 4 probes from cosmid 1-11 detecting RFLPs. Using all four probes, the PIC of the D12S6 locus is 0.73. To obtain the probe, cut cosmid DNA with BamHI and isolate the 11.6 kb fragment. To label the probe without removal of agarose, see BRL Focus 8:2 (spring 86). Prehybridize labeled probe with excess total human DNA. |
