Restriction digests of the clone give the following sizes (kb): BamHI--9.2; BglII--9.2; EcoRI--6.2, 1.65, 1.15. Cannot be used for the generation of gene knockout animals because the HPRT "minigene" does not function in embryonic stem cells. Can be used, by exploitation of LEU2 selection, for homologous recombination-based insertion into YACs, thus providing one approach to screening YAC libraries. The targets can be repetititive or unique sequences. Enables subsequent transfer into mammalian cells by conferring G418 resistance. Mammalian gene targeting vector designed for replacement or insertional recombination. Suitable for "hit and run" strategies and positive-negative enrichment of homologous integrations.
