The insert contains a DdeI site and a RsaI site. Restriction digests of the clone give the following sizes (kb): PstI--2.7, 0.64; EcoRI--3.3; HindIII--3.3. This construct does not make a fusion protein recognized by MAb PTY1. The insert includes coding sequences for the carboxy terminal portion of the coat protein gene, 3' untranslated sequences (166 nucleotides) and poly(A) (19 nucleotides). The 3' non-coding region is A + T rich (61%) and shorter than previously reported potyvirus sequences. There was no polyadenylation signal observed. |
