The insert contains internal AvaI, XbaI, HincII and HindIII sites. Restriction digests of the clone give the following sizes (kb): BamHI--2.9, 1.2; EcoRI--4.1; HindIII--3.3, 0.84; PstI--4.1; XbaI--4.0, 0.13. Linearize the plasmid with SalI for in vitro transcription. This is a full-length clone of virus segment S9. The cDNA ends were tailored to terminal BamHI sites before cloning into the BamHI site of pGEM-1. |
