Restriction digests of the clone give the following sizes (kb): NotI--2.4, 2.0; EcoRI/BamHI--3.0, 1.5; EcoRI/HindIII--3.0, 1.5; HindIII--4.5. Available for research purposes only. Plasmid containing an oriT mobilization cassette that can be inserted into general purpose vectors (ATCC 77278, 77279) for conjugal transfer of cloned inserts into the chromosome of P. aeruginosa. The oriT-containing fragment can be excised more precisely with an EcoRI/BamHI double digest. The NotI digest excises a fragment of approximately 1.9 kb that includes short segments of the pHSS21 vector. The NotI cassette includes these features: pHSS21 sequence - EcoRI - oriT - PstI - BamHI - XbaI - SalI - PstI - SphI - HindIII - pHSS21 sequence. |
