The insert contains an internal AccI site. Restriction digests of the clone give the following sizes (kb): BamHI--2.9, 1.6; EcoRI--4.5; HindIII--4.5; PstI--4.5; XbaI--4.5. Linearize the plasmid with SalI for in vitro transcription. This is a full-length clone of virus segment S7. The cDNA ends were tailored to terminal BamHI sites before cloning into the BamHI site of pGEM-1. |
