Restriction digests of the clone give the following sizes (kb): EcoRI/XbaI--4.5, 0.9; HindIII--5.4, EcoRI--5.4. This plasmid may be difficult to isolate and digest. If this problem occurs, increase the final concentration of ampicillin in the growth mediun to 1 mg/ml. Transformants secrete protein, inducible by threonine and repressible by glucose. |
