The EcoRI/BamHI fragment contains the following restriction sites (in nucleotides relative to the published sequence): XbaI--208, PstI--348, 455, 1038, 1103; NdeI--650, 823; PvuII--1639, EcoRI--2221. The coding sequences are contained in a EcoRV/BamHI fragment of about 3.25 kb which has been sequenced. Restriction digests of the clone give the following sizes (kb): HindIII/BamHI--5.0, 4.1; HindIII--9.2; BamHI--9.2; PstI--5.8, 1.15; 1.07; 0.64; EcoRI--5.2, 4.1; SalI--9.2. p3IT retains transforming activity which is inactivated by cleavage with EcoRI but not BamHI, HindIII, or EcoRV. Introns within the clone hybridize to highly repetitive mouse sequences. |
