PvuI and SmaI are unique restriction sites. Restriction digests of the clone give the following sizes (kb): BamHI--5.0, 3.0, 2.6; EcoRI--4.9, 3.4, 1.8, 0.4. The insert contains cDNA sequences from just before the initiator ATG (nucleotide 150-152) to nucleotide 7422 (200 bp downstream from the terminator at nucleotide 7201). The insert detects a single mRNA of about 9 kb from human liver. This was constructed by ligating several independently isolated cDNA clones and adding a SalI linker at nucleotide 145. |
