Restriction digests of the clone give the following sizes (kb): BamHI--13.4, 3.1; EcoRI--16.5; HindIII--9.8, 3.9, 1.6, 1.0.
The GenBank accession number for the adh insert is M15394.
The vector contains oriV from RSF1010, mob sequences from RSF1010 and pSUP205, and a Zymomonas mobilis promoter upstream of the BamHI cloning site.
Constructed by cloning a blunted EcoRI/SalI fragment containing both pdc and adhB into the SmaI site of pUC19. The BamHI fragment from this construct was cloned into the vector, such that both genes are controlled by the Z. mobilis promoter.
