The insert contains a PstI site located 1.5 kb from the 5' end. A PstI digest therefore yields the vector (3.8 kb) plus two insert fragments (1.5 and 0.8 kb). There is also a BamHI site at position 354. Restriction digests of the clone give the following sizes (kb): PstI--3.8, 1.6, 0.83; BamHI--5.0, 1.0; EcoRI--4.8, 1.3. This contains the complete coding sequence plus 5' and 3' untranslated regions. |
