Restriction digests of the clone give the following sizes (kb): BamHI--7.2; AvaI--7.2; PstI--7.2; EcoRI--7.2; HindIII--4.4, 2.8. Y-227 hybridizes to a BssHII fragment of 2.7 Mb also detected by Y-280. Y-227 hybridizes to a NotI fragment of 3 Mb which is not detected by Y-280. In HindIII digests, Y-227 hybridizes to a 2.8 kb Y-specific fragment and a smaller non-Y fragment. Each is single copy. The Y-specific band was detected in 8 of 11 46,XX males, but not in 2 46,XX hermaphrodites or 1 46,XY gonadal dysgenesis patient. Isolated from a flow-sorted human Y chromosome library enriched for DNA from the Y euchromatic portion. |
