Restriction digests of the clone give the following sizes (kb): BamHI--4.7, 1.4; EcoRI/BstEII--5.8, 0.3; PstI--4.7, 0.65, 0.35 (doublet); SacI--5.0, 0.7, 0.4; PvuII--2.5, 1.4, 1.3, 0.6, 0.25. Digestion with EcoRI+BstEII produces fragments of 5.75 and 0.35 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes the following mutant form: K32E, D33N, A34T, K38T, L39E, and residues 3-10 were replaced by the corresponding sequence from factor IX. |
