BamHI digestion gives a 2.0 kb fragment containing the VL exon, a 5.3 kb fragment containing the V MOPC-21, J2-J5, and Ckappa exons, a larger fragment about 7.5 kb, and 3 smaller fragments. Restriction digests of the clone give the following sizes (kb): EcoRI--16.0, 4.4; AvaI--13.0, 4.0, 3.7; BglI/XhoI--15.0, 2.4, 2.15; EcoRV--8.8, 6.2, 3.7, 1.0. A BglI/XhoI double digest gives a 14.8 kb fragment containing some pBR322 sequences plus all the exons, and smaller XhoI/BglI and BglI fragments. The insert is an EcoRI fragment from the MOPC-21 kappa gene which was cloned into Charon 4A (M21B1). The insert includes 5' flanking regions, VL, V MOPC-21, J2-J5, Ckappa, and approximately 8.2 kb 3' flanking sequence. |
