Restriction digests of the clone give the following sizes (kb): EcoRI--5.7, 0.66; HindIII--6.0; PstI--5.4, 0.37; BamHI--6.0; EcoRI/HindIII--2.95, 2.5, 0.62; MspI--2.3, 1.2, 0.45, 0.39, 0.29, 0.25; consistent with deposited DNA. Codominant segregation of the MspI RFLP was observed in 48 three-generation families. The insert appears to be chimeric, with the large MspI fragment detecting the polymorphism on chromosome 18. A fragment adjacent to the EcoRI side of the polylinker has been mapped to chromosome 9. Enzyme(s) not detecting polymorphism: RsaI, PstI, PvuII and BglII. |
