| Applications |
TK1 cells have a unique phenotype with high levels of the lymphocyte integrin, alpha 4, beta 7. TK1 cells are useful for studies of alpha 4, beta 7 function and regulation. |
| Storage Conditions |
liquid nitrogen vapor phase |
| Derivation |
TK1 is a T-cell lymphoma cell line originated by E. C. Butcher and I. L. Weissman from a spontaneous thymic lymphoma/leukemia that developed in an AKR/Cum mouse.

| Antigen Expression |
CD8 +; alpha4, beta7 integrin + |
| Comments |
They bind constitutively to the murine mucosal vascular addressin, MAdCAM-1.
TK1 cells have a unique phenotype with high levels of the lymphocyte integrin, alpha 4, beta 7.
They bind constitutively to the murine mucosal vascular addressin, MAdCAM-1. Ref Berlin C, et al. Alpha 4 beta 7 integrin mediates lymphocyte binding to the mucosal vascular addressin MAdCAM-1. Cell 74: 185-195, 1993. PubMed: 7687523
The cells are CD8 positive; they display approximately 1% of L-selectin of that expressed by normal lymph node cells. |
| Complete Growth Medium |
RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, and 1.0 mM sodium pyruvate and supplemented with 0.1 mM non-essential amino acids, 0.05 mM 2-mercaptoethanol, 90%; fetal bovine serum, 10%
|
| Subculturing |
Cultures can be maintained by addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in fresh medium at 1 to 2 x 105 viable cells/mL. Do not allow the cell concentration to exceed 2 x 106 cells/mL.
Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density). |
| Cryopreservation |
Complete growth medium supplemented with 5% (v/v) DMSO. Cell culture tested DMSO is available as ATCC Catalog No. 4-X. |
| Culture Conditions |
Temperature: 37°C
Atmosphere: Air, 95%; Carbon dioxide (CO2), 5% |
